Intended Use:
The Biopanda Human Chorionic Gonadotropin (hCG) ELISA test kit provides a means for the quantitative determination of hCG in human serum.
Principle of the test:
The test kit is designed in the format of a double antibody sandwich ELISA. In this test system, hCG present in samples binds onto wells of the microtiter plate coated with anti-hCG antibody. Bound hCG is then detected by using a Horseradish Peroxidise (HRP)-conjugated anti-hCG monoclonal antibody and visualised by adding the substrate TMB which gives a blue coloured product. This blue colour can be measured spectrophotometrically at 450 nm following the addition of an acidic stop solution which turns it yellow. The absorbance at 450 nm is a measure of the concentration of hCG in the test sample since the quantity of bound HRP varies directly with the hCG concentration which can be interpolated from a standard curve constructed using standards.
Materials and Components:
- Antibody-coated microtiter wells.
- Reference standards,0,5,20,50,150,300 mIU/mL, liquid standard, ready for use.
- Zero buffer, 12 mL
- Enzyme Conjugate Reagent, 18 mL
- TMB Substrate, 12 mL.
- Wash Buffer Concentrate(50X),15mL
- Stop Solution , 12 mL.
Storage:
Unopened test kits should be stored at 2-8°C upon receipt and the microtiter plate should be kept in a sealed bag with desiccants to minimize exposure to damp air. Opened test kits will remain stable until the expiration date shown, provided it is stored as described above. The shelf life is at least 2 years from manufacture when stored at 2-8°C. A microtiter plate reader with a bandwidth of 10 nm or less and an optical density range of 0-3 O.D. at 450 nm wavelength is acceptable for use in absorbance measurement.
